Connaître la biodiversité des bactéries et des champignons dans les atmosphères de travail est devenu essentiel pour mieux appréhender l'exposition aux bioaérosols et réduire le risque d'exposition. La mise en culture est la méthode la plus utilisée pour identifier et quantifier les bioaérosols, mais n'identifie que 1 à 10% des micro-organismes. D'autres méthodes basées sur la biologie moléculaire, comme le séquençage à haut débit (HTS), permettent d'étudier la quasi-totalité des microorganismes d'un échantillon d'aérosol. Le HTS nécessite une analyse bioinformatique pour identifier et quantifier la communauté microbienne. Plusieurs outils bioinformatiques existent, mais aucune information n'est disponible concernant leurs similitudes ou leurs différences. Dans ce travail, nous avons comparé deux outils bioinformatiques largement utilisés, Mothur et FROGS, sur des échantillons d'aérosols d'une usine de tri de déchets. Les deux outils apportent des résultats similaires pour l'identification des phylums les plus abondants. Ce travail mérite d'être approfondi au niveau du genre et pourquoi pas en intégrant d'autres outils de bioinformatique.

Abstract

Knowing about biodiversity of bacteria and fungi in workplace atmospheres has become essential towards being able to amin the risk of exposure. Culture is the most method used to identify and quantify bioaerosols but only identifies 1 to 10% of microorganisms. Others method based on molecular biology like high-throughput sequencing (HTS) make it possible to study almost all microorganisms in an aerosol sample. The HTS required bioinformatics analysis to identify and quantify microbial community. Several bioinformatics tools existed, but no information is available concerning their similarities or their differences. In this work, we compared two widely used bioinformatics tools, Mothur and FROGS, on aerosols samples of waste sorting plant. Both tools presented a high degree of agreement at identifying the most abundant phylums. This work deserves to be deepened at the genus level and why not by integrating other bioinformatics tools.
