Identification and quantification of bioaerosols in a cheese factory with a molecular biology approach

Résumé

La communauté microbienne des bioaérosols d'une fromagerie a été étudiée à l'aide de techniques de biologie moléculaire dans le cadre d'une démarche de prévention. Les bactéries et moisissures ont été quantifiées par PCR quantitative et identifiées grâce au séquençage haut débit, dans six zones d'activités de la fromagerie. La prédominance du genre fongique Penicillium et du genre bactérien Afipia a été observée. Les résultats indiquent aussi que la quantité de moisissures est nettement supérieure à la référence intérieure lors des activités de soins apportées aux fromages et que la composition de la communauté microbienne est différente en fonction de l'activité. La composition des bioaérosols en fromagerie est peu documentée ou essentiellement via une approche de cultivabilité. Ce travail présente donc une approche globale de caractérisation des bioaérosols et permet d'enrichir les connaissances sur ce type d'atmosphère professionnelle.

Abstract

The microbial community of bioaerosols from a cheese factory has been studied using molecular biology techniques as part of a prevention approach. Bacteria and fungi were quantified by PCR and identified with high-throughput sequencing, in six working areas of the dairy. The predominance of the fungal genus Penicillium and the bacterial genus Afipia has been observed. The results indicate that fungi concentration is significantly higher than the internal reference during cheese care activities and that the composition of the microbial community is different depending on the activity. The composition of bioaerosols in dairy is poorly documented or often via a culture approach, so this study allows for a global approach.
